Abstract

Purpose

This paper seeks to optimize a multiplex PCR in order to detect the incidence of Salmonella and Escherichia coli (E. coli) in chicken carcasses, eliminating a pre‐culture enrichment step and the pathogen isolation.

Design/methodology/approach

A total of 30 chicken rinse carcasses were analysed by standard microbiological methods, and the isolates were identified by biochemical and serological tests. The results were compared with those obtained by a multiplex PCR using validated primers targeting for invA and lamB genes of Salmonella and E. coli, respectively.

Findings

Microbiological analysis showed the prevalence of Salmonella in 14 out of 30 chicken carcasses. The same rinse samples were also analysed by multiplex PCR, which allowed the simultaneous detection of both bacteria directly from the chicken rinse water microbial community.

Originality/value

The optimized mPCR detected enterobacteria directly from the rinse samples, a complex matrix food, in one workday. There was 100 per cent agreement of the conventional microbiological analysis with those results obtained by multiplex PCR.

Abstract

Purpose

This study aims to evaluate the effects of betanin on AMP-activated protein kinase (AMPK), Sirtuin1 (SIRT1) and Sirtuin6 (SIRT6) gene expression as well as the tumour necrosis factor alpha (TNF-α) and interleukin 6 (IL-6) cytokine release in peripheral blood mononuclear cells (PBMCs) of patients with coronary artery disease (CAD) and healthy controls.

Design/methodology/approach

PBMCs isolated from whole blood of 50 patients with CAD and 48 healthy subjects aged 45 to 60 years were treated with 10 and 20 µM of betanin for 24 h. Real-time polymerase chain reaction was performed to assess gene expression levels of AMPK, SIRT1 and SIRT6. The supernatants of the cultured cells were used to assess the IL-6 and TNF-α protein levels by ELISA.

Findings

Treatment with both doses of betanin significantly increased AMPK, SIRT1 and SIRT6 expression in PBMCs of CAD patients compared to control non-treated cells (p < 0.05). In PBMCs of healthy subjects, only treatment with high dose of betanin showed significant increase in AMPK (p = 0.007), SIRT1 (p = 0.013) and SIRT6 (p = 0.024) expression compared to control non-treated cells. Betanin (20 µM) also significantly decreased TNF-α and IL-6 concentrations in the culture supernatants of the CAD patients compared to control non-treated cells (p < 0.001).

Originality/value

Betanin could enhance AMPK, SIRT1 and SIRT6 gene expressions in PBMCs and represent a useful complementary treatment to reduce the proinflammatory status accompanied with CAD.

Abstract

Purpose

This research aims at studying the influence of a classroom’s inner environmental conditions on undergraduate students’ performance using an experimental methodology.

Design/methodology/approach

The Uchida-Kraepelin test (U-K test) was applied to measure the performance of a group of 47 students in a selected classroom that was arranged according to the following experimental conditions: air-conditioning on, and doors and windows closed (D1); doors and windows open, and air-conditioning off (D2); air-conditioning off, and doors and windows closed (D3). After completing the tests, questionnaires were distributed to evaluate the students’ assessment of each set of environmental conditions.

Findings

On-site measurements of humidity and carbon dioxide levels stress the importance of ensuring good natural ventilation through open doors and windows, independently of whether the air-conditioning system is operated or not. Also, the authors find that the students’ self-assessment regarding the inner environmental conditions for each studied set was entirely accurate, with set D3 being assessed as the worst. The U-K test scores for each environmental set did not show statistically significant differences, which means that, in the studied conditions, the student’s performance in the tests was not affected by the inner environmental conditions.

Practical implications

There is a direct relationship between the building’s indoor conditions and an occupant’s health. Factors such as poor maintenance, bad indoor environmental quality (IEQ) and building age will worsen the building’s condition and negatively impact the occupant’s health. Educational buildings with poor IEQ can reduce the concentration and performance of occupants.

Social implications

School is an important place to help students grow in their various capabilities. They spend approximately 30% of their daily lives in schools for their educational activities. Since most of their activities are performed indoors, indoor environmental attributes, such as light, heat, air and sound, should be maintained as required. In general, schools are not thermally comfortable. The extreme thermal environment of classrooms affects students’ concentration. Thermal discomfort may also cause irritation. In addition to reduced concentration, such an environment could also cause tiredness, sluggishness and health problem.

Originality/value

Despite the importance of the issue, scientific investigations of the correlations between students’ performance and the quality of scholar buildings’ inner environmental conditions are still relatively recent. In this context, this research further explores the effect of a classroom’s different environmental inner conditions on the performance of undergraduate students at a university in São Paulo/Brazil.

Abstract

Purpose

The civil construction industry has vital importance to Brazil's economy. However, this sector is also responsible for the environmental impacts. Governments have been taking measures aiming to mitigate these impacts. Among these, the elaboration and implementation of civil construction solid waste management plans can be highlighted. However, these plans still lack standardizations and tools for their evaluation. Environmental indicators proposal for construction solid waste management plans assessment is presented to verify the adhesion of these to environmental laws, technical standards and green building certification systems recommendations.

Design/methodology/approach

The construction solid waste management plans of three construction works were evaluated by the proposed indicators to verify the procedures related, generating, in the end, a scale between 0 and 5. After that, plans were compared with each other.

Findings

The proposed indicators have made possible the evaluation of the environmental practices performed for three different construction works. By the proposed indicators, the environmental practices were compared to technical standards and legislation suggested procedures.

Practical implications

As a contribution, the evaluation proposal presented may help the construction industry as well as the public authority to evaluate the construction solid waste management plans currently elaborated, so that these can offer a quality improvement and more effective environmental measures.

Originality/value

Methodologies that guide the evaluation of construction solid waste management plans can be beneficial for the construction companies, which can improve the quality of the plans elaborated internally and verify the effectiveness of the plans elaborated by specialized consultancies. In general, most of the construction solid waste management plans are prepared with the purpose of only complying with the legislation, more specifically of the National Council for the Environment, Resolution 307/2002.

Historic districts are unique with its urban fabric, which hosts various activities and land uses. Such districts in the city center of cities are very active with many different users. Each activity attracts different users. Thus, urban mobility is very essential issue for these districts to function properly. Recent urban mobility needs are very different to those in former times, when these districts were built. Therefore, these districts are no longer compatible for contemporary urban mobility, thus, there are many problems facing these districts in reference to the urban mobility.

Zamalek, as one of the most important colonial district in the city center of Cairo, suffers from deteriorated urban mobility situation. This deterioration is mainly due to concentration of activities and densification. This paper is analyzing the existing condition in Zamalek in terms of urban mobility. This, also, includes the urban planning analysis in relation to the urban mobility. The current problems are very challenging, thus innovative concepts shall be implemented to ensure sustainable urban mobility in Zamalek. This paper recommends a set of action plans localized for the current condition in Zamalek and based on successful practices in other cities.

Abstract

Purpose

Holistic renovation of historic and heritage buildings involving different stakeholders has the potential to reduce environmental impact. Climate change concerns are emphasizing environmental issues of cultural built heritage leading to new policies, guidelines and methods dealing with the challenge on how to lessen the environmental impact of built heritage without damaging its cultural significance. The purpose of this paper is to review existing international and New Zealand holistic guidelines for renovation of historic and heritage buildings.

Design/methodology/approach

A systematic review is used to identify international projects, methods and criteria within the holistic approach. Secondly, the New Zealand context is explored and compared with best international practices.

Findings

For instance, in New Zealand one increasingly important issue is the many vacant heritage and historic buildings in provincial town centres that need action to deal with building code seismic changes. Upgrades and adaptive reuse are opportunities to make them more sustainable and climate change resilient. However, the lack of national holistic guidelines regarding the challenge of reducing environmental impact whilst keeping the heritage values is a critical gap which urgently needs to be resolved. The need is further increased within the context of the recently passed Zero Carbon Act 2019, which aims for national zero carbon emissions by 2050.

Originality/value

These shared responsibilities for conserving historic and heritage buildings to maintain life in provincial towns on one hand, and the need to reduce greenhouse gas (GHG) emissions on the other, have the potential to contribute to a sustainable development of cities and communities.

Abstract

Purpose

Heritage buildings are consistently impacted by technical and pathological issues associated with their maintenance and conservation such as diminish of building's authenticity and damaging environmental impact. This paper aims to evaluate the environmental maintenance impact (EMI) of the Singgora roof tiles repair in heritage buildings. The EMI is an evaluation upon embodied carbon expenditure during maintenance phase, thus important in repair efficiency appraisal.

Design/methodology/approach

Calculation procedures within selected boundaries of life cycle assessment (LCA) and arbitrary period enabled evaluation of the EMI of Singgora roof tiles repair in heritage buildings during the maintenance phase.

Findings

Evaluation of the EMI could be appreciated as a carbon LCA of Singgora roof tiles repair and has been recognised in embodied carbon expenditure reduction in the form of CO2 emissions mitigation. Importantly, the evaluation underpins decision-making for heritage buildings repair.

Practical implications

EMI evaluation encompasses all building types and forms, thus comprehends the associated applied methodologies. Moreover, the evaluation reflects the emerging environmental challenges of sustaining resilient buildings globally.

Social implications

EMI evaluation highlights options that may be adopted in repair. Indirectly, this implicates heritage building preservation and place's identity protection. Significantly, the evaluation supports environmentally focused conservation and promotes a sustainable repair approach.

Originality/value

EMI evaluation of this paper may devoted to the holistic understanding of the complex relations between Singgora roof materials and their environmental performance. Meanwhile, the application of a carbon LCA had dictated integration of multidisciplinary of heritage buildings maintenance and conservation.

Abstract

Purpose

This study attempts to identify and analyse the barriers to implementing a reverse logistics (RL) system in Indian Construction, Real estate, Infrastructure and Project (CRIP) sectors and present a structured model to identify interdependencies among them.

Design/methodology/approach

The barriers to implementing RL in CRIP sectors in India were identified using a Delphi study. The interdependencies were identified using Interpretive Structural Modeling (ISM). Further, using the Matriced' Impacts Croisés Multiplication Appliquée à un Classement (MICMAC) analysis, the barriers were classified on the basis of their driving power and interdependencies.

Findings

The study has identified ten barriers that can hamper the application of an RL system in CRIP sectors. The finding of the ISM model shows that macro level barriers such as lack of awareness of reverse logistics, insufficient government policies and unavailability of standard codes stimulate each other and also drive all other barriers. The organization-specific barriers operating at the strategic/tactical level, namely, company's rigid mechanism, lack of awareness of economic profits, inadequate company’s organizational policies and lack of training, reluctance from stakeholders, scarcity of resources and finance from company are found at the intermediate level of hierarchy and they can be influenced by the barriers at the lower level and influence the barriers on the and higher levels. The operational level barrier namely “Inadequate Information Technology system” is at the top of the hierarchy and can be driven by all the barriers at the lower level.

Research limitations/implications

The present findings are based on the opinions of experts only from Indian CRIP sectors so the results may require to be validated in other contexts.

Practical implications

The structural model presenting the interdependencies will be a guide for the CRIP supply chain professionals in understanding and ranking the barriers they may face while implementing the RL system.

Originality/value

The study contributes to the existing literature by providing a set of barriers and their interdependencies faced during the implementation of an RL system implementation in CRIP sectors. It is one of the first studies which identifies barriers applicable to the CRIP firms in India and models their inter-dependencies. Additionally Consequently, these firms can make a move forward towards a circular economy by overcoming these interlinked barriers.

Purpose

Kefir is a traditional fermented dairy beverage that has numerous health benefits due to the presence of bacteria and yeasts in an exopolysaccharide matrix. This study aims to isolate and identify beneficial microorganisms and evaluate the antimicrobial activity of kefir beverage against two important food-borne pathogens including Salmonella Typhimurium and Listeria monocytogenes.

Design/methodology/approach

Microorganisms were identified by the polymerase chain reaction with specific primers, and antimicrobial activity was evaluated by the disk diffusion method.

Findings

The following microorganisms were identified as natural inhabitants of the kefir grains: Leuconostoc lactis, Lactococcus lactis subspecies lactis, Streptococcus cremoris, Enterococcus faecalis, Enterococcus faecium, Lactobacillus helveticus, Leuconostoc mesenteroides, Lactobacillus fermentum, Lactobacillus reuteri, Lactobacillus plantarum, Lactobacillus casei, Bifidobacterium langum, Saccharomyces cerevisiae and Pichia fermentas. Also, the results obtained from the disk diffusion method showed the inhibitory effect of kefir milk on Salmonella Typhimurium and Listeria monocytogenes with an inhibitory average diameter of 8.3 ± 4–9.1 ± 2.8 and 3.4 ± 3–6.6 ± 3 mm, respectively.

Originality/value

The results of this study showed that Iranian traditional kefir beverage contained different species of lactic acid bacteria and yeasts and has antimicrobial activity against two important food-borne pathogens, Salmonella Typhimurium and Listeria monocytogenes, which the highest inhibitory effect was observed against Salmonella Typhimurium.
